Anticoagulant properties of glycosaminoglycans derived from cultured endothelial cells

چکیده
Anticoagulant properties of culture medium conditioned by human umbilical vein endothelial cells were investigated. In the presence of conditioned medium, both prothrombin time and activated partial thromboplastin time of normal plasma were prolonged by 4 and 10 seconds respectively compared with those in the presence of nonconditioned medium. The prologation of coagulation time was not affected by the presence or absence of serum in the culture medium, indicating that endothelial cells release the anticoagulant activity into the culture medium. The concentrated serum-free conditioned medium, not only directly inhibited Factor Xa activity but also enhanced the inactivation of Xa activity by antithrombin III. On the other hand, acidic glycosaminoglycans (GAG) isolated from conditioned culture medium showed Xa inhibition only in the presence of antithrombin II. These results suggest that anticoagulant properties of endothelial conditioned medium are, at least in part, attributed to GAG(s) released by these cells.

Cultured endothelial cells produce heparinlike inhibitor of smooth muscle cell growth
Using cultured cells from bovine and rat aortas, we have examined the possibility that endothelial cells might regulate the growth of vascular smooth muscle cells. Conditioned medium from confluent bovine aortic endothelial cells inhibited the proliferation of growth-arrested smooth muscle cells. Conditioned medium from exponential endothelial cells, and from exponential or confluent smooth mus...
متن کاملEffect of Purification of Human Adipose-derived Mesenchymal Stem Cells on the Expression of vWF Cell Factor Under Chemical and Mechanical Conditions
Introduction: Human adipose-derived mesenchymal stem cells (hADSCs) are easily accessible in the body, and under appropriate conditions, they can be directed toward various phenotypes. Therefore, hADSCs have been considered as a potential cell source for tissue engineering applications. hADSCs are able to differentiate into endothelial cells which covers the interior surface of vessels, in vi...

Heparin was isolated from whole clam tissue Marcia opima . Purification of multimilligram quantities of heparin from this tissue sample permitted a thorough examination of its anticoagulant activity characterization, which was partially purified by fractionation by anion exchange chromatography using DEAE cellulose column. The isolated crude and partially purified fractionated sample showed me...
متن کاملTumor Associated Mesenchymal Stromal Cells Show Higher Immunosuppressive and Angiogenic Properties Compared to Adipose Derived MSCs
Background: Differentiation, migratory properties and availability of Mesenchymal Stromal Cells (MSC) have become an important part of biomedical research. However, the functional heterogeneity of cells derived from different tissues has hampered providing definitive phenotypic markers for these cells. Objective: To characterize and compare the phenotype and cytokines of adipose derived MSCs (...
متن کامل3D study of capillary network derived from human cord blood mesenchymal stem cells and differentiated into endothelial cell with VEGFR2 protein expression
New blood forming vessels are produced by differentiation of mesodermal precursor cells to angioblasts that become endothelial cells (ECs) which in turn give rise to primitive capillary network. Human cord blood (HCB) contains large subsets of mononuclear cells (MNCs) that can be differentiated into endothelial-like cells in vitro. Human mononuclear progenitor cells were purified from fresh umb...
